Paraoxonase gene polymorphism analysis in patients with diabetes mellitus

Diabetes Mellitus (DM) is one of the most important disease in the worldwide because of consisting serious complications of mortality and morbidity. It take important place in the world due to increasing incidence in medicine. Determining of the gene polymorphism in childhood, Diabetes Mellitus that associated with multifactorial etiology will lead to understanding of the genetic background of the disease. In this study, we aimed to determine whether the possible or impossible relationship between Paraoxonase1 55 (PON1 55) and 192 polymorphisms in pediatric patients with DM and to determine correlation between fasting glucose, HbA1c, cholesterol and high density lipoprotein (HDL) in among different genotype groups. We examined PON1 55 and PON1 192 polymorphisms in 75 children who were diagnosed with DM and 54 healty controls whose age and gender were matched similarly with mean age 3-15. After DNA extraction we determined the genotypes of the individuals by PCR-RFLP methods. Also, blood samples such as HDL, LDL, total cholesterol and glycose were taken from patient group and control group and only blood samples for HbA1c level were taken from patient group. Statistical analysis was performed by SSPS 16.0 (Chicago ILL USA). Qualitative data were compared between groups by the chi-square test and t-test was used for the analysis of numerical data. There is no significant difference between patient and healthy control group (p>0,05) in examining polymorphisms. Weak relationship was identified between levels of HDL, total cholesterol, glucose and polymorphisms of PON1 55 and PON1 192. PON1 55 polymorphism frequency was statistically significantly higher in patient group with levels higher than %6,6 of HbA1c (p<0,05). There was no significant association between PON1 192 polymorphism and HbA1c level (p>0,05). In conclusion, although no statistically significant relationship was found between the paraoxanase gene poymorphism and DM, this study is important in terms of being first study in pediatric age group about the relation between paraoxanase gene polymorphisms and Diabetes Mellitus according to our current literature knowledges.
