Abstract (EN)

Epilepsy is a neurological disorder that is widespread globally and affects people of all ages, characterized by its chronic nature. It can be caused by many factors and is marked by recurring seizures. Excessive or prolonged alcohol use can lead to persistent seizures. Understanding the physiological events that occur in the brain and muscle tissue following alcohol consumption is crucial for developing preventive measures against this condition. Therefore, this thesis aims to investigate changes in oxidative stress parameters in epilepsy-like seizures induced by pentilentetrazole in zebrafish following alcohol use. In this study, zebrafish were exposed to ethanol at doses of 1%, 1.5%, and 3% daily for 1 hour. After 28 days of treatment, epileptic seizures were induced by administering 15 mM pentilentetrazole (PTZ). Subsequently, brain and muscle tissues were collected, and changes in oxidative stress parameters such as malondialdehyde (MDA), total glutathione (GSH), superoxide dismutase (SOD), and catalase (CAT) as well as antioxidant enzyme activities were investigated. In the study, a statistically significant increase in MDA levels was observed in the groups exposed to alcohol, PTZ, and alcohol+PTZ. Meanwhile, there was a dose-dependent decrease in SOD, CAT, and GSH levels, with significant reductions detected. The most pronounced adverse effects were observed in the group exposed to 3% alcohol and PTZ. Based on the results, it can be concluded that both the alcohol and alcohol+PTZ groups experienced degradation of the antioxidant mechanisms and the development of oxidative stress. Key Words: Zebrafish, Danio rerio, epilepsy, pentylenetetrazole, oxidative stress, antioxidant enzymes
