Abstract (EN)

This study was carried out to construct the first genetic linkage map of pistachio using ISSR, SRAP and AFLP molecular marker techniques. Two separate genetic linkage maps have been constructed by applying the strategy of `double pseudo-testcross? mapping with 92 F1 hybrid individuals of a cross progeny which were derived from crossing between `Siirt? (Pistacia vera L.) cultivar and monoecious `PA-18? (Pistacia atlantica Desf.) genotype. In this research, 19 markers segregated by analysing 16 ISSR primers, 9 of which were identified in `Siirt? cultivar and 10 in `PA-18? genotype. Besides, from 27 SRAP primer combinations, 96 markers segregated, 49 of which were identified in `Siirt? cultivar, and 47 belonged to `PA-18? genotype. In addition to these, 206 markers segregated from 27 AFLP primer combinations, of which were identified in the `Siirt? cultivar and in the `PA-18? genotype as 107 and 99 markers, respectively. In accordance with this study, a total of 165 markers (9 ISSRs, 49 SRAPs, 107 AFLPs) belonged to the map of `Siirt? cultivar, of which 87% 1:1 and 13% 3:1 segregated with an average marker number of 9.70. Besides this, in the map of `PA-18? genotype, of 156 markers (10 ISSRs, 47 SRAPs, 99 AFLPs) with a mean marker number of 8.21, 86% and 14% found to have 1:1 and 3:1 segregation, respectively. In this study, `Siirt? genetic map had 17 linkage groups spannig 1237.3 cM length with a mean marker density of 7.50 cM and `PA-18? genetic map comprised 19 linkage groups covering 1337.5 cM with a mean marker density of 8.57 cM. This study has the first basis of genetic linkage maps of pistachio for molecular breeding. In the future mapping studies, more hybrid progenies should be analyzed and more dominant markers should be used together with co-dominant markers like SSR to obtain better genome coverage for a better saturated high density maps.
