Studies on interleukin 10 gene promoter (2849 A-G) and angiotensin converting enzyme intron 16 insertion/deletion polymorphisms and genotype distributions in preeclampsia patients

Preeclampsia is a multifactorial disease characterized by hypertension and proteinuria. Placental, maternal, immune and genetic factors are thought to play an important role in its pathophysiology. Besides, the relation between preeclampsia and many environmental factors also genetic structures which affect the onset of preeclampsia was analyzed and the existence of many candidate genes was reported. However, the effect of these genes on preeclampsia, the interaction with each other and environmental factors remains to be unclear.In this study, 126 preeclamptic and 116 normotensive pregnant women was researched in terms of the relationship between preclampsia and interleukin-10 (IL-10) gene 2849 promotor and angiotensin converting enzyme (ACE) gene intron 16 insertion/deletion (I/D) polymorphisms. Moreover, the distribution of genotype and allele frequencies belonging to these polymorphisms in normal and preeclamptic women was also evaluated.According to our results, there was significant difference in terms of genotype and allel frequencies between preeclamptic and normotensive pregnant women. For IL-10 gene 2849 promotor polymorphism, AA genotype was found significantly higher in preeclamptic women than normotensive pregnants (p=0,030), but there was no relation between GG genotype and preeclampsia. Codominant, dominant and recessive models was appplied in ACE gene I/D polymorphim and for codominant model, DD genotype was found significantly higher in preeclamptic women than normotensive pregnants (p=0,016), but there was no relation between II genotype and preeclampsia. In dominant model (DD, II+DI) again there is a significant relation between DD genotype and preeclampsia (p=0,006) but no relation in recessives (p=0,977). For IL-10 gene 2849 promotor polymorphism, G allele frequency was 85.7% in preeclamptic group while it was 87.5% in controls (p=0,564). For ACE gene intron 16 I/D polymorphism, D allele frequency was 64.6% in patients while it was 56.1% in controls (p=0,062). There is no significant difference between preeclamptic and normotensive pregnants considering the allele frequency
