Preparation and in-vitro testing of progesterone-loaded nanocomposites based on polyurethane, polyacrylonitrile, and chitosan containing boron compounds

Abstract (EN)

Within the scope of this thesis; firstly, progesterone (Prg) loaded and unloaded polyurethanes containing disodium octaborate (DOT) (DOTPU, Prg-DOTPU-1, Prg-DOTPU-2) were obtained that can be used in estrus control in animals in veterinary medicine. It was determined their structures with FT-IR, thermal tests with DSC and TGA, their morphology with scanning electron microscope (SEM). The percentage swelling ratios of the obtained polyurethanes were determined. Drug release studies of progesterone loaded polyurethanes containing DOT were performed in buffer solution at 25 oC and 37 oC (In-vitro). Progesterone release values were measured in UV at 248 nm wavelength. When compared with progesterone loaded commercial implants, it was determined that the obtained progesterone loaded polyurethane implants released for a longer time than the commercial implants Another study; Prg loaded nanofibers (Prg-PAN10, Prg-PAN5, Prg-PAN/DOT, Prg-PAN/BN) were obtained and their structures were illuminated with FT-IR. In addition other work; Composites containing different amounts of DOT (PE/DOT, PE/PAN/Chi/DOT, PE/DOT/PAN/Chi/BN, PP/DOT, PP/PAN/Chi/DOT, PP/DOT/PAN/Chi/BN) and composites containing different amounts of h-BN (PE/BN, PE/PAN/Chi/BN, PE/DOT/PAN/Chi/BN, PP/PAN/Chi/BN, PP/DOT/PAN/Chi/BN) were produced in micro-extruder. For comparison, composites without h-BN/DOT compounds (PE/Chi, PE/PAN/Chi, PP/Chi and PP/PAN/Chi) were also synthesized in micro-extruder. All the obtained composites were investigated structures with FT-IR and their thermal degradation with TGA. As a result, firstly new prg-loaded polymeric materials were obtained and their drug releases were evaluated. In addition, when the mechanical tests of these new polyurethanes and composite materials without progesterone are performed, they can be used for various industrial applications other than veterinary medicine.
