Role of LNCRNA MEG3 on RB1 related genes in prostate cancer

Abstract (EN)

Prostate cancer is a heterogeneous disease that is ranked as the second leading cause of cancer-related deaths in males worldwide. Progression of prostate cancer is relatively slower than other cancer types. However, it still accounts for more than 10% of cancer-related mortality in males. Therefore, achieving a comprehensive understanding of the basic molecular mechanisms in developing prostate cancer can be helpful in developing effective treatment strategies. Recently, increasing evidence has shown that long non-coding RNAs (lncRNAs) provide new insights into the understanding of prostate cancer pathogenesis. MEG3 (Maternally Expressed Gene 3) is a tumour suppressor lncRNA and it has 12 different isoform including MEG3b. RB1 (Retinoblastoma Transcriptional Corepressor 1) is also another tumor suppressor gene which loss of activation is the cause of many cancers including prostate cancer and its expression level is affected MEG3 expression by DNMT1. Both of MEG3 and RB1 have role in main cellular processes and they share some common targets including p53, Mdm2. In this study, MEG3 gene expression profile was performed by using 30 different cancer and normal cell lines and 20 different types of normal human tissue. MEG3 and MEG3b was overexpressed by plasmid transfection in PC-3 and DU 145 prostate cancer cells, where gene expression was the lowest in cancer cells. Overexpressed MEG3 and MEG3b altered the expression of RB1 and its targets while suppressing proliferation, colony formation and migration of prostate cancer cells.
