Investigation of the effect of propolis on oxidative stress in acetaminophen-induced hepatotoxide model in rats

Acetaminophen is used as an analgesic and antipyretic drug due to it spain-relieving and fever-reducing effect. High doses of acetaminophen may cause hepatotoxicity. This study was conducted to evaluate the curative effect of propolis, one of the bee products, against acetaminophen-induced liver dysfunction and oxidative stress. Forty-one 2.5-month-old male Wistar-Albino rats were used in thes tudy, and they were divided into 5 groups: goup 1; control group, group 2; propolis, group 3; acetaminophen, group 4; propolis+acetaminophen, group 5; acetaminophen+propolis applied groups were formed. In the propolis group, 200 mg/kg/day propolis was dissolved in ethanol and administered by gavage for 7 days. The acetaminophen group was administered 2 g/kg by gavage as a single dose of acetaminophen. Propolis+acetaminophen administration was started 7 days before acetaminophen administration andc ontinued for 7 days. Acetaminophen+propolis administration was started immediately after the administration of acetaminophen and continued for 7 days. At the end of the applications, malondialdehyde (MDA), reducted glutathione (GSH) levels, catalase (CAT), glutathione-S-transferase (GST), glutathione peroxidase (GSH-Px) activities were measured in blood and liver tissue samples. When the acetaminophen administered group was compared with the control group, a significant increase in blood and liver MDA levels, a significant decrease in GSH levels, CAT, GST (p<0.001) and GSH-Px (p<0.05, p<0.001) activities were observed. When the groups given acetaminophen+propolis and propolis+acetaminophen were compared with the group given acetaminophen, a significant decrease in MDA level and a statistical increase in CAT, GST and GSH-Px activities in GSH level were foundand it approached the control group. (p<0.001). It was determined that the improvement was more pronounced in the group treated with propolis+acetaminophen, but GST activity in the blood could not be measured because it was at an unreadable level. As a result, based on the obtained biochemical data; It can be said that propolis has protective and therapeutic effects against acetaminophen-induced liver dysfunction and oxidative damage.
