Investigatıon of the efficacy of amniotic fluid in the treatment of experimental alkaline burns generated experimentally in rats

It can initiate ulceration in the cornea by allowing epithelial passage through the eye of microorganisms when it is caused by direct or indirect causes of corneal epithelial integrity. Corneal alkaline burns may also cause perforation of the ulcer by disturbing the integrity of the cornea. When treating corneal alkaline burns, pay attention to the following points. The chemical agent is removed from the medium and the re-epithelialization is regenerated. There are many medical treatment options available for this. Some of them are amniotic membrane and amniotic fluid applications. Due to the numerous growth factors that amnion possesses, the epithelium accelerates, fibroblastic activitiy increases and thus it contributes to the collagen synthesis and has a pain and inflammation reducing effect. This study was conducted to investigate the efficacy of amniotic fluid on experimental burned alkaline burns in the cornea. A total of 30 female albina rats were used in the study. The rats were randomly divided into 3 equal groups. In the first and second groups under anesthesia, alkaline burns were formed, waiting of the 2NOH solution, which was saturated on 3 mm fitler paper, for 40 seconds on the cornea in the right eyes. No action was taken in the third group. 3 times a day for 20 days, two drops of the cow's amniotic fluid was dripped in the first group of rats, when two drops of saline was dripped in the second and third group of rats. During this time ocular findings were evaluated by using indirect ophthalmoscope and, clinical improvement in corneal was also evaluated by using fluroscein and rose bengal painting. At the end of the 20th day, rats in all groups were euthanized. The epithelial healing in cornea was also studied histopathologically and biochemically. At the end of the clinical examinations, tear secretion, break time of tear (TBUT) and findings obtained from the result of fluroscein and rose bengal painting, were more satisfying in the second group where amniotic fluid applied, the results obtained in the rats in the control group were worse than the first group. In the histopathological and biochemical evaluations, epithelial healing in cornea of the first group of rats was revealed histopathologically, while the increase in levels of GSH-Px, MDA and GSH in the cornea was revealed biochemically. As a result, it has been concluded that amniotic fluid was effective in the corneal alkaline burns and therefore it would be useful to use it in addition to the medical or operative treatment.
