Abstract (EN)

SUMMARYThe Effect of Xylol and Formaline Inhalation to Testicular Tissue in RatsIn this study, the immunohystochemical and histometrical changes of testicular tissuesobtained from the rats which their mothers were subjected to xylol and formaline from thebegining of pregnancy, after the birth and immediately after puberty were evaluated.Nigty six male Sprague Dawley?s rats were chosen as biological material. The diversegroups were arranged in order to their ages and the duration of exposure. One group of rats?mothers exposed to xylol and formaline gases during pregnancy and then the baby rat exposedto xylol and formaline gases until they were five weeks of age. One group of rats wereexposed to xylol and formaline gases from birth to eigth weeks of age, n done group of ratswere exposed to xylol and formaline gases from puberty for eigth weeks. Each group of ratsexposed to diverse gas formulations such as xylol (300 ppm), formaline (6 ppm) andksilol+formaline (150 ppm+3 ppm) combination for eight hours each day for eight weekslong. Control groups for each groups of rats, exposed to gases, were taken to compare thedifferences. After the tests, the groups, five weeks old, eight weeks old and 18 weeks old,respectively were weighed and their testis weights were determined.The blocks were prepared for colorations and then two coloration methods were appliedto the transversal sections; triple coloration methods to define histometric changes and streptavidin-biotine complex (sABC) to show relaxin like factor (RLF). RLF?s tissue densities weredefined with light microscope (Leica DMLB) and the area of tubulus and the height ofepithelium of tubulus were defined with image analyze system which was linked tomicroscobe (Leica Q Win Standart).In this study, Sertoli and Leydig cells were defined in each groups of rat testes. From theage of eight weeks spermatids were defined well. In the group of rats, 18 weeks old, had clearspermatozoons inside the lumen of tubules.In control groups of those diverse ages, Leydig cells were determined to be positive inRLF when immunohistochemically checked. In the others, coloration to determine RLFshowed that there was no influence in the group which were exposed to xylol gas but in thegroups which were exposed to formaline gas showed a loss in density of coloration in eigthand 18 weeks old groups.The groups which were exposed xylol+formaline gases, whom are eight and 18 weeksold showed that there was no colaration for RLF.The weight of testes was not affected from each chemical gases inhalation; testicularweights increased with increasing age and body weight.Statistically, there was no influence of age, body weight, chemical substance andchemical substance-age interactions on the area of tubulus.Epithelium height was affected by formaline and xylol+formaline inhalations, there is aclear loss of height in these groups but there was no influence of xylol. Regarding each groupsof rats, five weeks old ones had higher epithelium than the others.Consequently, formaline and xylol+formaline combinations had negative influences onLeydig cells (RLF) and those tubular epithelium of rat testises.Key words; Histometrical, tubulus seminiferous contortus, relaxin-like factor (RLF),xylol, formaline, rat
