Immunohistochemical and ELISA methods screening of endocan molecule in renal cell carcinoma tissues

Renal cell carcinoma (RCC) is a group of malign arising from the renal epithelium. It is a tumor that is diagnosed between the ages of 55-60 on average and is usually seen in adults. Endocan has been used as a blood-based and tissue-based biomarker for various cancers and inflammation. We will evaluate the tissue endocan levels of renal cell cancer subtypes immunohistochemically and biochemically. We will investigate whether it can be used in the diagnosis and follow-up of patients diagnosed with renal cell cancer (RCC). The files of the patients who underwent nephrectomy at Fırat University Medical Faculty Hospital were reviewed retrospectively, and the paraffin blocks of 140 patients whose pathology results were reported as RCC were examined. The patients were divided into RCC subgroups and the tissues obtained were stained with endocan and analyzed histochemically and biochemically by ELISA method. The patients in our study were divided into groups as Normal Kidney Tissue, Chromophobe Cell RCC, Papillary Cell RCC, Clear Cell RCC Grade-1, Clear Cell RCC Grade-2, Clear Cell RCC Grade-3 and Clear Cell RCC Grade-4. Endocaan levels were found to be significantly higher in grade 2, 3 and 4 subtypes of chromophobic RCC and clear cell renal cancer, among these patient groups, when evaluated both by immunohistochemistry and by ELISA method. In conclusion, endocan is an endothelial-derived molecule and is also secreted from normal tissues. It has been shown to increase more than normal in diseases such as endocan sepsis, ARDS, endometrium cancer, renal cell cancer, liver cancer, brain glioblastoma, breast cancer. In some renal cell cancer subtypes, endocan is more prominent. More extensive studies are needed to determine the use of endocan in RCC.
