Transcriptional investigation of the effect of extracts from Rheum ribes L. medicinal plant on obesity and insulin resistance in 3T3-L1 cell line

Abstract (EN)
Obesity is a low-grade inflammatory disease that manifests itself in developed countries and causes major health problems. Excessive accumulation of adipose tissue, which occurs by increasing the size or number of adipocytes, is associated with a number of diseases such as Type II diabetes, cancer, hypertension, insulin resistance. Genetic (metabolic rate) and environmental factors (unhealthy diet and physical inactivity) are positively correlated with obesity. Compounds of plant origin; It is very important because of its various biological effects such as antidiabetes, anticancer, antiobesity, antihypertension, antioxidant and anti-inflammatory. Rheum ribes has been used as a source of medicinal drugs in Turkey and the world since ancient times. Rheum ribes plant is traditionally used in the treatment of diabetes, hypertension, high cholesterol, constipation, psoriasis, ulcers, diarrhea, wounds and cuts. The blood sugar-lowering activity of the plant has been demonstrated as a result of studies. It has been included in the literature that this plant is consumed against obesity and digestive problems, but there are not many studies. In this study, it was aimed to elucidate the molecular mechanism of Rheum ribes plant associated with obesity and insulin resistance. After the plant extracts were given to the differentiated 3T3-L1 adipocyte cells, total RNA isolation was performed. After going to cDNA analysis from RNA, qPCR analyzes were performed with the cDNA samples obtained. It is known that control groups and plant extract treated cells are associated with obesity (Dgat1, Lpl and Fasn), inflammation (Mcp1 and Il-6), insulin resistance (Irs-1) and extracellular matrix (Col V) by Real Time PCR method. Trials were set up with primers designed via NCBI. Normalization was achieved by using 18S as the housekeeping gene. As a result of the study, a decrease in the expressions of Dgat1, Lpl, Fasn, Col V, Il-6 and Mcp1 was observed and these changes were found to be statistically significant.
