Abstract (EN)
Testicular torsion or spermatic cord torsion ,especially seen in children and young men, ends up needing a surgical intervention. among urological cases. It is one of the most important reasons why men infertility has to be observed and early diagnosis can protect the testis from irreversible damage. In the studies about torsion/ reperfusion damage,there is a wide variety of vitamins,medicines and antioxidant that are used for protective measures. EGCG takes place in flavanoids, which form eighty percent of green tea. It is known that EGCG in green tea is related with PKA and PKC which are signalization patways in testicular androjen production, known to produces testesterone. Cape propolis is an active component of honey bee. CAPE propolis which is similar to flavanoids has been shown to have antioxidant ,anti-inflammatory, antiviral immüne-modulator effects. mTOR is one of the basic kinase enzymes, which coordinates many critical cell functions. mTOR is a member of PI3K/AKT/mTOR signal patways, from the serin-threonin kinase family. mTOR effects cell growth, cell proliferation ,synthesis and transcription of proteins, angiogenesis, apoptosis and autophagy. In this study our aim is to evaluate the importance of PI3K /AKT/mTOR signal message ways and protection effects of EGCG and CAPE, which occur among antioxidant agents biologically on testis damage. Structure damage which occurring after reperfizyon in young adult Wistar Albino rats will evaluate using indirect Immunocytochemstry method. Histopatological examination will be done using Johnson's score and all cells which included in the process of spermatogenesis and seminifer tubulus, Sertoli cells and interstitial Leydig cells will be analysed.In biochemical blood analysis,will be evaluated by measuring levels of MDA,Katalaz,8-OHG. It will be investigated whether appliance of EGCG and CAPE have protective effects on testis torsion/reperfusion damage or not by histopatological evalution under the microscope. DNA damage will be defined by using the TUNEL method. Defining colored positive cells as percentage, results will be evaluated using the ANOVA statistical test, relatively among groups. Evaluation will be made by an indirect immunohistochemical method with the aim of showing the effects of extrinsic and intrinsic ways in the apoptatic process with primer antibodies which take place in PI3K/ AKT/ mTOR signal pathways immunocytochemstry wise,with Caspes 8 and Caspes 9. H-score will be calculated by defining immunochemstry coloring scores as mild (1),moderate(2),strong(3),very strong(4). The results will be evoluated relatively using the ANOVA statistic test by defining the total cell numbers mulptiplying among h-scores groups. At the end of this study, we will be evaluated the effects of EGCG and CAPE on testis torsion damage which is one of the reasons of infertility at a molecular level. The importance of synergic effects will be stated by observing the protective effects of treatment, when antioxidants are used one by one or all together.
