Effect of methylphenidate hydrochloride on orthodontic tooth movement and root resorption in rats

Abstract (EN)
The aim of this study is to examine the effect of the drug on tooth movement and root resorption when orthodontic treatment is initiated in individuals using methylphenidate. A total of 30 wistar albino rats were used for this purpose. 30 rats were divided into 3 main groups and 3 main groups into 2 subgroups. The first group, the SF group; It is divided into 2 subgroups as SF-30 and SF-44. 5 ml/kg saline solution daily for 30 days was given to 5 rats in the SF-30 group by gavage and then sacrificed. 5 ml/kg saline solution was given daily by gavage to 5 rats in the SF-44 group for 44 days, and 50 grams of orthodontic force was applied to the right first molar teeth of the rat on the 31st day of the experiment. After 14 days of orthodontic force application, 5 rats in the SF-44 group were sacrificed. The second group, the MSD group; It is divided into 2 subgroups as MSD-30 and MSD-44. 5 rats in MSD-30 group were given 3 mg/kg methylphenidate solution daily for 30 days by gavage and then sacrificed. 5 rats in the MSD-44 group were given 3 mg/kg methylphenidate solution daily by gavage for 44 days, and 50 grams of orthodontic force was applied to the right first molar teeth of the rat on the 31st day of the experiment. Following orthodontic force application for 14 days, 5 rats in the MSD-44 group were sacrificed. The third group, the MAD group; It is divided into 2 subgroups as MAD-30 and MAD-44. 5 rats in the MAD-30 group were given 3 mg/kg between the 1st and 8th days, 6 mg/kg between the 9th and 16th days, 9 mg/kg between the 17th and 24th days, and between the 25th and 30th days. whereas 12 mg/kg methylphenidate was given by gavage and then sacrificed. 5 rats in MAD-44 group 3 mg/kg between 1st and 8th days, 6 mg/kg between 9th and 16th days, 9 mg/kg between 17th and 24th days, between 25th and 44th days and 12 mg/kg methylphenidate was given by gavage. On the 31st day of the experiment, 50 grams of orthodontic force was applied to the right first molar teeth of 5 rats in the MAD-44 group and after 14 days of orthodontic force application, 5 rats in the MAD-44 group were sacrificed. Orthodontic tooth movement and root resorption in all groups; Micro Computed Tomography was examined by histomorphometric and immunohistochemical evaluation methods. Orthodontic tooth movement (Micro-CT and Histological Examination) was higher in the "MAD-44" group compared to the other study groups. In our study, no statistically significant difference was found between the groups for root resorption volume
