Relationship of Schizosaccharomyces pombe sdj 1 gene with cellular protection mechanisms

Parkinson's disease (PD) is a neurodegenerative disease without a known cure. It is little known how and why neurons fail in PD. In this process, it is emphasized that 6 genes have an effect on PD formation: α-synuclein, Parkin, PTEN-stimulating putative kinase1 PTEK1, DJ-1, leucine-rich repeat kinase2 (LRRK2) and ATP13A2. In this study, the sdjl gene of Schizosaccharomyces pombe, which is known to play a role in various cellular processes such as detoxification of dicarbonyl compounds, autophagy and oxidative stress response, was discussed. In this study, we aimed to investigate the effect of sdj1 gene, which is the ortholog of DJ-1 gene in S. pombe, on the cellular responses related to stress in endoplasmic reticulum at biochemical level. In the study process, firstly, growth curve of S. pombe sdj1  and parental strains for the application of tunicamycin was prepared. According to this, after applying 1μg / μL tunicamycin 1, 4 and 24 hours at the 15th hour of the culture in parental and 17th hour of the culture in sdj1  , the susceptibility of sdj1  to tunicamycin compared to S. pombe parental race, determined. Intracellular oxidation analysis and cell morphology were examined by fluorescence microscopy with tunicamycin application for 4 hours. Finally, the apoptosis response of S. pombe sdj1  against tunicamycin administration was investigated in comparison with the parental race. As a result, in the absence of S. pombe sdj1 gene (sdj1  ), increase in sensitivity and intracellular ROT level (1.5 times in S. pombe parental race, 2.5 times in S. pombe sdj1  race) is associated with sdj1 gene stress. However, it was determined that no apoptosis was associated with tunicamycin.
