Determination of micropropagation and rooting performance of selected apricot genotypes under in vitro drought stress conditions

In the thesis study, the micropropagation performances of 5 genotypes, obtained after selection studies on apricots within the scope of PRIMA (Project No: 118O855) project, under in vitro drought stress conditions were investigated. In addition, the effects of different nutrient media (MS - Murashige and Skoog Medium, ½ MS and WPM - Woody Plant Medium) and IBA (Indole Butyric Acid) concentrations (1 and 2 mg/l) on rooting were investigated. MS nutrient medium containing different concentrations of PEG8000 (Polyethylene Glycol) (0, 1, 2, 4 and 6%) was used in in vitro drought experiments. 1 mg/l BAP was added to the nutrient media. At the end of the study, the highest micropropagation coefficient was obtained in the Aldeniz (2.76) genotype in the PEG-free (control) group. The highest plant height was obtained in Hatay 1 (1.54) genotype in the PEG-free (control) group. In rooting experiments, the highest number of roots was obtained in Hatay 1 (4.50) genotype in ½ MS nutrient medium containing 1 mg/l IBA. The maximum root length was obtained in ½ MS (1.37 cm) nutrient medium containing 2 mg/l IBA. As a result of in vitro drought studies, it has been determined that plants survive and micropropagated in all nutrient media. However, it was determined that plant growth was negatively affected at increasing PEG concentrations.
