Abstract (EN)
In this study, 3 cellulase positive anaerobic bacteria Clostridium sp. were isolated from samples taken from solid waste landfill in Osmaniye province as defined. The enzyme with the highest cellulase activity strain 4 was produced from these bacteria and the enzyme produced was characterized. The enzyme was determined to be an alkaline thermophile enzyme with an optimum operating temperature of 60 ° C and an optimum operating pH of 8. The enzyme released 6.95 mg / mL reducing sugar after 12 hours in 1% CMC substrate solution under optimum conditions. Biogas production using isolated cellulase produced 3710 ° C in total in 12 days and 5110 mL of biogas was produced. In the study carried out at 55 ºC temperature, 5640 mL biogas production was realized in the control group with no enzyme in 7050 mL and in the control group with no enzyme in 12 days. The fact that the enzyme produced is thermophile has shown an effect in the production of biogas, resulting in a 20% increase in the amount of gas produced. In this study, alkaline thermophile cellulose was isolated from Clostridium sp. from the landfill and it can be said that it is an enzyme that can be used in thermophile operated biogas systems.
