Abstract (EN)

Bioconjugates which are synthesized by reaction of synthetic peptides and polyelectrolytes are biologically important. Their application on biological fields has great potential. There are lots of research related to functional biopolymer systems in last years. These researchs are about synthesis, characterization and application properties of biologically important polyelectrolytes. This work is aimed to microwave assisted bioconjugate synthesis of synthetic peptides and polyelectrolytes. Viral peptides, which are used in this work, are synthesised with microwave assisted solid phase peptide synthesis (SPPS) method in research laboratory. These peptides are characterized with LC-ESI-MS system. Also amino acid analysis of one peptide sequence is done. Polyconidine is one of the polyelectrolytes used in this work. Firstly monomer of polyconidine is synthesised, after homopolymer of conidine is synthesised with different initiator types. Polyconidine is biodegradable polymer and it is an ideal polymer for preparation of bioconjugates. Vinyl pyrrolidone/acrylic acid copolymer (VP/AA) is another polymer which is used in this work. VP/AA has biodegradable properties, too.Bioconjugate synthesis of peptide and polyelectrolytes with microwave assisted by HBTU/HOBt activators in organic media is a novel application. This new method is more effective than classical method which in water media with carbodiimid activation. The new methods advantages are, time consuming and precipitation properties. Peptides which synthesised in this work selected from viral capsid part of three different diseases (FMDV, Influenza, Melanoma). Synthesis of synthetic vaccine can be possible conjugation of polyelectrolytes and with peptides which choosed for a specified reason. Immunological research with synthesized bioconjugates is processed by our research group. Results of immunological analysis is hopefulness.Key words: bioconjugation, LC-ESI-MS, microwave, polyelectrolyte, polyconidine, solid phase peptide synthesis
