Evaluation of some parameters before pcr in molecular tests of local grape varieties in Siirt province

In this research, grape varieties grown in Siirt and its districts have been determineted. From this varieties, leaf samples have been taken. In this context, DNA concentrations and purity quantities of leaf samples taken from 21 grape varieties which take part in this research have been defined. After obtained outcomes are analyzed, it has been defined as smallest quantity of DNA concentration of local varieties 121.5385 (ng/ul) in Cevzen variety; highest quantity 1006.9231 (ng/ul) in Emiri variety. Also, purity quantities belonged to varieties which take part in this research, it has been detected as smallest 0.8759 (A260/A80) in Emiri variety; smallest quantity 1.1765 (A260/A280) in Kışmır variety. Last of all, patterns belonged to genetic materials obtained from this research have been put under protection. These samples will be an important source for forward researches which are about this topic. Also; it has been thought to make researches about these samples on analyzed at molecular level, genetic mapped, defined and detected their similarity relationships.
