The investigation of the effects of curcumin on expression of sirts in cisplatin-induced nephrotoxicity in rats

In this study we aimed to determine the effects of curcumin on renal expressinon of sirtuins (SIRT) and Nicotinamide phosphoribosyl transferase NAMPT in cisplatin induced nephrotoxicity. We used 28 wistar albino male rat (8 wk-old, between 200-215 g) in this study. We separated the rats into 4 groups (n:7 for each group) as; Control group (n:7): Standard diet and saline administration intraperitoneal with equal volume of cisplatin (1ml/kg/day). Cisplatin group (n:7): cisplatin administered in %0, 9 saline solution on the third day of study. Curcumin group (n:7): curcumin administration 2 days before and continued 8 days after saline administration. Cisplatin + curcumin group (n:7): cisplatin+ isotonic saline solution with equal volume and curcumin administration 2 days before and continued 8 days after cisplatin administration. At the end of study, rats were decapitated and serum and renal tissues were obtained for biochemical and histopathological analysis. Serum urea, creatinine and tissue MDA, SIRT1, SIRT3, SIRT4, NAMPT levels were evaluated. Renal tissues were also histologically evaluated. Serum urea, creatinin and tissue MDA levels increased and; SIRT1-3-4 and NAMPT levels decreased in cisplatin group in comparison with control group. In cisplatin + curcumin group; serum urea, creatinin and tissue MDA levels were significiantly lower; SIRT1-3-4 and NAMPT levels were significiantly higher in comparison with cisplatin group. Histopathologic findings such as vacualisation, tubular necrosis, tubular atrophy and inflammation were relatively low in cisplatin + curcumin group. In conclusion, we determined that curcumin reduces lipid peroxidation; increases cirtuin and NAMPT levels. According to the results of our study, protective effects of curcumin on cisplatin induced nephrotoxicity depends on the increasing expression of cirtuins and NAMPT.
