Arli OT. Synthesis, Characterization and In Vitro Applications of Cisplatin-Loaded Micellar Casein Nanoparticles. Istanbul University, Institute of Health Science, Department of Biophysics. Master of Science Thesis. Istanbul. 2021. In this study, cisplatin, one of the most common and effective anti-cancer agents used in cancer treatment, was encapsulated with sodium caseinate without using any cross-linking agents. Casein nanoparticles were synthesized in the recombined micellar structure with the help of citrate, phosphate and calcium ions and by simple coacervation. The purity and molecular weight of the subunits of the synthesized nanoparticles were determined by SDS PAGE analysis. Effective diameters of 196.4 nm and polydispersity were found to be 0.226 by DLS analysis. SEM and TEM analyzes were used to observe spherical micelles compatible with the Holt Model. Cisplatin-casein binding was determined by ATR-FT-IR analysis. Pt was determined by ICP-MS analysis and it was determined that casein nanoparticles encapsulated cisplastin with an efficiency of 94.8%. The cytotoxicity of cisplatin-loaded casein nanoparticles in 4 different cell lines (MCF-7, A-375, 3T3 and HUVEC) was examined with the MTT test and it was observed that while they showed low cytotoxicity in healthy cell lines in vitro, they were more cytotoxic in cancer cell lines. In order to determine whether cisplatin reaches the cells, ICP-MS analysis was performed by taking the supernatant of the cells treated with certain doses before the MTT test in MCF-7 and HUVEC cell lines, and it was determined that cisplatin reached the cells at a high rate in these cell lines.Key Words: Nanoparticles, Drug Delivery Systems, Caseins, Cisplatin, Cancer The present work was supported by the Research Fund of Istanbul University.
