Abstract (EN)
Scleroderma is a rarely seen chronic collagen tissue disease which has been fist described in 18 th century. It is indicated that the mutations in BMPR 2 (bone morphogenetic protein receptor type II) gene affected the course of disease.In this study; the determination of BMPR 2 gene expression levels via Real Time PCR and its association with the clinical course in scleroderma patients were revealed.20 scleroderma diagnosed and 29 healthy individuals were added to this study and their blood samples were taken at Çukurova University Medicine Faculty, Department of Internal Diseases, Clinic of Rheumatology. Performing the transformation of c-DNA complementer to m-RNA isolated from leucocyte cells, DNA amplification was carried on via Real Time PCR. To determine the expression level in BMPR 2 gene, beta actin gene was selected as the reference gene.In control group average levels of gene expression were established as 0, 0023875883 (approximately %0,2), in patient group that was 0,0007295126 (approximately %0,07). Expression level of the patients decreased approximately %0,13 when it was compared with the control group PAH (pulmonary arterial hypertension) improvement was seen in 4 patients (%20) from 20 patients added to this study and the gene expression level of this patients was found to 0,001 (approximately %0,1).The gene expression level of patients increased approximately %0.03 in proportion to other patients examined in this study.Consequently, in scleroderma patients BMPR2 gene expression level showed changes from person to person but decreased by comparison to control, in all the PAH patients that was found high by comparison to other patients.
