Abstract (EN)

Western-style diet has become an important danger in societies with the developing industrialization and the increase of fast food culture. Consumption of a high-fat diet is effective on the initiation and progression of the carcinogenesis process in the colon, which is one of the most effective organs in the composition of feces, which provides the absorption and release of water and electrolytes during the nutrition and digestion process, contains a large variety of important bacteria and is one of the most effective organs in the composition of feces. For all the type of diet consumed, it has been proven that the nutrition of the individual both in the maturation period and in the maternal period affects the epithelial tissue and microbiota in the colon. As a result of high fat diet, colonic mucosal barrier dysfunction occurs it causes luminal antigens and bacteria to enter the lamina propria, triggering immune cell-mediated mucosal inflammation. Consequently, the production of reactive oxygen species increases in colon epithelial cells. Carcinogenesis occurs as a result of genomic instability in the colon with the effect of genetic and environmental factors. In the literature, there are studies to explain the effects of mutations in the DNA repair mechanism and various signaling pathways on the development of colon carcinogenesis. Gamma-H2AX, a DNA damage marker, and RAD51, a double-stranded DNA repair marker, are prominent markers in determining the process of carcinogenesis. In this study, maternal (21 days of gestation+21 days of lactation) and maturation (100 days) rats were fed a low fiber and high fat western diet. We aimed to show DNA damage and repair with immunofluorescent staining method, morphological changes with hematoxylin-eosin staining and oxidative stress change with a commercial kit, which are the parameters that are indicative of carcinogenesis formation in the colon of exposure to this diet. In our results, no statistically significant results were observed in RAD51 and H2AX expressions in total oxidant and antioxidant stress conditions, while a tendency to the formation of abnormal crypt foci was observed in histological sections taken from the colon tissue of the groups fed with high-fat maturation period. In line with these results, it is predicted that the possible negative effects of high-fat diet on the colon at different periods of development are observed and may contribute to future studies.
