Abstract (EN)

Molecular markers have recently become an important part of plant breeding programs. Accurate and reliable results can be achieved in marker aided breeding studies. Although SNP and SSR markers have been widely preferred, InDel (insertion-deletion) markers developed by next generation sequencing technologies are also an important alternative for breeding approaches. With these markers, differences can be monitored in agarose gels in any laboratory where basic molecular devices are available. In addition, high polymorphism and inter-species transferring makes the InDel markers are highly preferable for molecular assisted selection, genetic diversity and genetic mapping studies. In order to make progress in sesame studies, introducing different marker types is crucial for breeding process. In this thesis, InDel markers were developed by using the data obtained from next generation sequencing of 95 sesame genotypes originating from various regions of the world. Differing from the reference genome, 785 indel regions over 2 bp were detected and 86 regions over 8 bp were identified which were distinguishable on agarose gel. The 38 of the identified 86 InDel regions were used for primer design and all of these markers were successfully amplified with use of bulk DNAs. The selected markers were also demonstrated the genetic diversity of sesame germplasm including 32 genotypes. The developed InDel markers are expected to contribute to molecular-based sesame breeding studies worldwide.
