Synthesis of bis (1,2,3-triazole n-oxide) based dinuclear coordination compounds: Characterization and investigation of DNA interactions

In this thesis, a 1,2,3-triazole N-oxide ligand was synthesized from the reaction of dimeric starting material obtained from the reaction of 2,4-dihydroxyacetophenone with 1,3-dibromo propane and 4-chloro isonitrosophenylhydrazine. Ni (II), Cu (II), Co (II), Zn (II) complexes were synthesized with this ligand and the structures of the compounds were characterized by elemental analysis, FTIR, 1H-NMR, 13C NMR. DNA binding and cleavage activities of the obtained ligands and complexes were investigated depending on time and concentration. In addition, DNA cleavage mechanisms were elucidated by cleavage studies in the presence of various radical scavenging and cavity binding agents. In addition, the binding interactions of all compounds with CT-DNA were determined by UV absorption spectroscopy. Binding results show that ligands and complexes bind to DNA by intercalation or partial intercalation. With a binding constant of 3.00×104 M-1, the Cu (II) complex showed the best DNA binding activity. In addition to the effects of complex concentration and incubation time on DNA cleavage reaction, cleavage mechanisms were also investigated with DNA cleavage experiments. It was observed that the ligand showed high cleavage activity in the presence of hydrogen peroxide and the Cu (II) complex showed the highest cleavage in all compounds at the same concentration. Mechanistic studies show that the species responsible for oxidative cleavage reactions are hydroxyl and singlet oxygen radicals.
