Investigation of Trametes versicolor and Phonerochaete chrysosporium BKM-F 1767 laccases; productive secration condations depanding on inducer and proper mediator for their effect on indigo dyes

Abstract (EN)

Main source of laccases, which exist widely in nature and have glycoprotein structure and molecular weights range from 50.000 to 130.000, are fungus. Since they form water as the final product, this enzyme group is called as environment-friendly and it has been taking increasing interest. This enzyme group, which has a wide application field from textile to paper industry as well as waste refinement processes and of which substrate specifity is quite wide, catalyses reactions proceeding with the electronic oxidation of the substrate and converting it into the corresponding reactive radical.So far, more than one hundred laccases were isolated and characterized from nature. After 1990 when it was realized that ABTS behaved as a laccase substrate mediator or improve the effect of the enzyme, usability of many other compounds as substrate for this purpose was investigated. Consequently, it was established that the mediator compounds can be changed depending on source of enzyme that is isolated and/or the substrate. Since development of proper laccase/mediator systems for such purpose will reduce foreign dependence in the related industries, it contributes cost reduction of the products.In this labour, excreting more laccases of (melon-watermelon peels and red pine cone) in which it was used various support materials of Phonerochaete chrysosporium and Trametes versicolor was elicited by stimulating them by solid phase culturing and various inducers. According to this, in two different concentrations of these inducers; The maximum laccases excreting of 50 µM 2,4-xilidine cone is on 26th day, 5000 µM 2,5-xilidine watermelon is on 26th day, 5000 µM 3,5-xilidine is on 12th day, 5 µM synthesized is on 26th day culturing. It was dispensed with the large scale production because it was not observed enough amount of protein and (enough) laccases activity of(in) P. chrysosporium. The KM value of T.versicolor was found as 0.120, the Vmax value was found as 20868,114 in pretestings.Besides redox mediator , can show catalytical effectiveness for decolorising the indigo colours used commonly in txtile industry, of enzymes (deoxyribonucleases) determination oriented studies were done. When we compare the mediators selected for indigo colour with MS and HBT (1-hidroksibenzotriazol), It was observed that they are not usually effective, but NHP, RF and MPP mediators are as effective as at least MS or HBT in decolorising of indigo carmine.Keywords: Fungus, inducer, laccase, mediator, indigo decolouration
