Development and characterization of buccal nanofiber formulation containing triamsinolone acetonide

Various ulcerative conditions may occur in the mucosal membrane lining the oral cavity. Corticosteroids are among the most frequently used drug classes in the treatment of such diseases. One of the commonly used agents in this group, triamcinolone acetonide, exhibits low oral bioavailability when administered via the oral route and therefore available on the market in topical formülations such as ointments and gels. However, these formulations are often rapidly cleared from the site of application due to saliva flow, swallowing reflex and eating/drinking activities, which limits their ability to maintain effective therapeutic concentrations. Within the scope of this thesis, nanofiber based formulations were developed using two different polymers as an alternative to the ointment based formulation currently available on the Turkish pharmaceutical market. The primary objective was to enhance the release properties of the drug and thus improve the effectiveness and practicality of treatment. The developed formulations were sıbjected to comprehensive characterization studies and in vitro drug release as well as ex vivo permeability studies demonstrated that triamcinolone acetonide was successfully released from the nanofiber matrices.
