Abstract (EN)
Colorectal Cancer (CRC) is one of deadly cancer; It is a multifactorial disease. There is a poor prognosis for patients who diagnosed late. Complete understanding of the pathogenesis at the molecular level could have significant implications concerning the detection and treatment of CRC. Moreover, establishing early non-invasive diagnostic biomarkers will aid the course of the disease treatment and overall survival. MicroRNAs are critical elements in the progression of cancer and are candidate biomarkers detectable in blood. We studied the expression level of circulating level of six miRNA, namely; miR-195, miR-92a, miR-126, miR-1247, miR-214, and miR-224, to correlate their level with CRC stages and compare the samples from patients and control group among the Turkish population. We performed qRT-PCR, to detect the expression level of miRNAs in 40 patients have colorectal cancer and 40 control individuals, for statical analysis, we used one-way ANOVA and Pearson correlation test. Our study revealed that miRNAs (miR-195, miR-224, miR-126-3p and miR-1247) are significantly downregulated in patient's serum compared with the control group with a fold change (-18, -6.4, -9 and -11, respectively), with a p-value less than 0.05. while miRNAs (miR-214 and miR-92a) are upregulated in cancer, having 6.6 and 35 fold-change orderly between the patients and control cases. Interestingly, there is a significant difference in expression level among different cancer stages. There is an insignificant difference in the expression level of miRNAs based on smoking, age and gender. However, mir-1247 was higher among the smoker group. The result indicates the possibility of using those miRNA as biomarkers in colorectal cancer.
