Özet (EN)
The research was conducted to determinate genetic diversity among ninety-one chickpea genotypes sampled from different region of Turkey and two chickpea cultivars using Inter Simple Sequence Repeats (ISSR) and Sequenced Related Amplified Polymorphism (SRAP) DNA molecular markers. To determine best polymorphic primers for ISSR and primer combination for SRAP, sixty-five ISSR primers and eighty-two SRAP primer combinations were tested on five chickpea genotypes and two chickpea cultivars. Based on this study, eleven ISSR primers and nine SRAP primer combinations, yielding strong and reproducible polymorphic bands, were selected for further analysis. Both DNA molecular marker techniques gave very low polymorphism in chickpea genotypes and cultivars. According to the Jaccard?s similarity index, similarity between studied chickpea genotypes and cultivars, ranged from a minimum of 0.64 to a maximum of 1.00, with average of 0.90 UPGMA dendrograms based on Jaccard similarities obtained from two datasets separated the chickpea genotypes and cultivars into two main groups. Cluster B, divided into two main sub-clusters: B1, containing 3 chickpea genotypes, and B2, containing 85 chickpea genotypes and 2 cultivars. Cluster A has only three genotypes. The end of this study, It was found that Turkish chickpea genotypes exhibited very narrow genetic diversity at DNA level.
