Abstract (EN)

This study aims to investigate the antioxidant activities and cytotoxic effects of methanol extracts obtained from Xanthoparmelia pulla (Ach.) O. Blanco, A. Crespo, Elix, D. Hawksw. & Lumbsch, a speciesdistributed in Turkey. The amounts of secondary metabolites for Xanthoparmelia pulla were determined using High-Performance Liquid Chromatography. According to the obtained results, the highest amount was found to be Salazinicacid (37.23 ± 9.21 mg/g extract). The total phenolic content (172.77 ± 10.50 mg GAE/g extract) and total flavonoid content (9.25 ± 1.32 mg CA/g extract) of Xanthoparmelia pulla extract were determined. DPPH (1,1-diphenyl-2-picrylhydrazyl) and ABTS [2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid)] methods were used for antioxidant activity evaluation. It was determined that Xanthoparmelia pulla extract had the highest DPPH scavenging activity, showing 87.41% inhibition at a concentration of 2 mg/mL. Similarly, thehighest ABTS scavenging activity was found to be equivalentto 2.29 ± 0.31 mmol/L Trolox at a concentration of 0.5 mg/mL. Cytotoxic activity assessment was performed on MCF-7 and MDA-MB-231 usingthe MTT assay. According to the results, Xanthoparmelia pulla extract exhibited 33.16% viability at a concentration of 125 μg/mL in the MCF-7 cellline, while 60.58% viability was observed at a concentration of 250 μg/mL in the MDA-MB-231 cell line.
