Abstract (EN)

Plant extracts with medicinal and therapeutic characteristics have grown in popularity owing to the severe side effects of synthetic drugs. The research aimed to evaluate the chemical composition of the leaves and stems of P. oleracea collected from three distinct areas (Iraq, Istanbul, and Çatalca) utilizing water and 70% methanol extracts. The phenolic content of the samples was compared, and their antioxidant activity was evaluated using the DPPH method. The antibacterial activities of the extracts were examined using the disc diffusion technique against two microorganisms, E. coli and S.aureus. Additionally, SDS-PAGE was used to visualize proteins in various samples and compare their total soluble protein content. The results showed that methanol extraction typically showed higher antioxidant activity, despite the geographical region or specific plant part. The methanol extract of Catalca leaves had the highest antioxidant activity with a value of 4514.23±591.01mg Trolox /100 g DW and a total phenolic content of 8.7±0.22mg GA/100 g. Additionally, Catalca leaves have a greater total soluble protein content. Regarding antibacterial activity, the Catalca leaves methanolic extract showed an inhibition zone (10.8mm-10.7mm) and Iraq leaves methanolic extract (9.2mm-11.1mm) against S. aureus. The results indicate that P. oleracea has active phytochemical substances that might potentially be used as antioxidants and for anti-inflammatory therapy.
