Is it beneficial to use bretschneider histidine-tryptophan-ketoglutarate solution before reperfusion after revascularization in rats with prolonged limb ischemia?

Abstract (EN)
The aim of this study was to determine the effect of the HTK solution on ischemia / reperfusion injury after acute onset of ischemia. Thirty two Wistar female rats were used in the study. These rats are divided into 4 groups; Control Group (Group 1, n= 8), surgical stress group (Group 2, n= 8), ischemia group (Group 3, n= 8) and HTK group (Group 4, n= 8). Only biopsy was performed in the control group, and biopsy was performed in the surgical stress group 2 hours after the right main iliac artery exploration with median laparotomy. The right common iliac artery of ischemia and HTK group animals were ligated after median laparotomy and 8 hours of ischemia followed by 2 hours of reperfusion was created. In the HTK group, 1 cc HTK solution was injected from the abdominal aorta immediately after the surgical knot was removed (40 cc/kg - 1/6). Biopsy was performed for histopathological examination of the distal half of Gastrocnemius-Soleus muscle group in all groups. The samples were histopathologically graded by light microscopy in terms of muscle fiber degeneration and leukocyte (PMNL) infiltration. When histopathological data obtained from all groups were analyzed statistically, a successful ischemia model was formed in rats (p= 0,001). However, no significant difference was found between the ischemia group given the HTK solution and the ischemia group not given the HTK solution (p= 0,195). A significant difference could not be found between HTK-administered ischemic group and non-HTG ischemic group, because it is possible that the organ-protective effect of HTK may be involved when given as a pre-ischemic, not post-ischemic. However, better pathological results in 25% of subjects with HTK suggest that the statistical significance could be related to the number of subjects. It is thought that more accurate results can be achieved by increasing the number of subjects. The absence of serious side effects of the HTK solution suggests that similar experiments can be applied to other mammalian animals (sheep, dogs, etc.).
