Abstract (EN)

In this study, the sediment sample used for the isolation of methylotrophic bacteria was supplied from the area where the Van Metropolitan Municipality Wastewater Enhancement Plant was opened. In this study, bacterial reproduction was observed in the isolation of methylotrophic bacteria only in a medium containing 1-5% methanol of one isolate. Sequence analysis of the 16S rRNA gene region was performed by determining the morphological, physiological and biochemical properties of this isolate called Halomonas S1 isolate. It was observed that only S1 isolate were able to produce only carbon from the isolates and 1% methanol in the feed as energy source. In order to obtain a more precise result, it was observed that S1 culture had the ability to use methanol as the only energy and carbon source after it was tested using NMR technique. The S1 isolate compared to the 1202 nucleotideregion in the 16S rDNA gene region phylogenetic analysis was found to be similar to Halomonas desiderata (NR 026274) with 16 nucleotide differences and 98,6% similarity. İt is also found that the database in the GenBank using BLAST(Basic Local Alignment Search Tool) programe is Halomonas desiderata with 100% similarity to the comparator isolate.
