Abstract (EN)

The aim of this study is to accomplish all the necessary steps and design a protocol for seed based in vitro clonal micropropagation of Vigna caracalla L. Verdc. The seeds provided from a commercial firm are incised and cultured in MS medium after exposed to different sterilization methods. The highest percentage of sterilization (%93.33) and germination (%93.33) are achieved when the seeds are soaked in %70 ethyl alcohol for 1 min, 0.1 % HgCl2 for 4 min and sterile distilled water for 7 hours. The seeds are cultured in different media compositions and culture vessels, the highest percentage of germination (%95.24) is determined in combinations of N6 medium and 250 mL Erlenmeyer flasks. Higher germination percentage (%85.71) was determined in MS medium solidified with gelrite compared to agar. Higher germination percentage (%80.00) was detected at 3500 lux light intensity in seeds cultured in WPM nutrient medium at different light intensities. When the shoot tips and node explants of in vitro seedlings are cultured for shoot regeneration in MS, WPM and N6 based medium including gelrite, the highest propagation coefficient (1.71) is achieved from shoot tip explants cultured in WPM medium. The highest propagation coefficient (1.48) is achieved in shoot tip explants cultured in MS medium including 1IBA+0,5BAP (HS4). In the experiments carried out to determine the effect of gelling agent, light intensity and explant type on shoot regeneration, the highest propagation coefficient (1.71) is achieved in node explants cultured in N6 (N-21) medium solidified with Duchefa agar and exposed to 4200 lux light intensity. Within the scope of this study, it was determined 16 clones that higher multiplication rate and without high hyperhydricity showed appropriate reactions in micropropagation experiments. The highest root regeneration (85.71%) is achieved from shoot explants cultured in N6 medium solidified with Fluka agar and exposed to 4200 lux light intensity. The rooted plantlets are acclimatized with 70% success percentage.
