The protective role of rejuveinix, A combination of vitamins, on kidney damage and inflammatory response in lipopolysaccharide-galactosamine-induced septic shock model

Sepsis, which causes multi-organ failure, is the most common cause of morbidity and mortality in intensive care units worldwide. In this study, we aimed to investigate the therapeutic effects of rejuveinix, which has anti-inflammatory, antioxidant and renal protective effects in sepsis and sepsis-related acute kidney injury. Our study was conducted after the approval of Fırat University "Animal Experiments Local Ethics Committee", and 40 male BALB-c mice (age: 8 weeks old) were randomly divided into 4 groups. Control, LPS+GaIN, LPS+GaIN+RJX 2.5 and LPS+GaIN+RJX 5 groups were composed. The LPS+GaIN group was put into lipopolysaccharide-galactosamine septic shock. LPS+GaIN+RJX 2.5 group was treated with RJX 2.5 ml/kg and LPS+GaIN+RJX 5 group with RJX 5 ml/kg. In our study, kidney function tests; BUN, creatinine, proinflammatory cytokines; TNF-alpha, IL-1ß, IL-6, TREM-1, antioxidant markers; MDA, anti-inflammatory cytokine in serum and kidney tissue; SIRT-1 levels were measured. The data of the group Control, LPS+GaIN, LPS+GaIN+RJX 2.5 and LPS+GaIN+RJX 5, according to TNF-alpha, IL-1ß, IL-6, TREM-1, SIRT-1, BUN, creatinine, serum and kidney MDA were made and statistically a significant difference was found between all groups when compared in terms of their levels (p<0.05). When the LPS+GaIN group was compared with the control group, TNF-α, IL-1ß, IL-6, TREM-1, BUN, creatinine, serum and kidney MDA levels were significantly increased, while SIRT-1 levels were significantly decreased (p<0.05). In the same parametres of LPS+GaIN+RJX 2.5 and LPS+GaIN+RJX 5 groups, opposite changes were found. These changes were more pronounced in the LPS+GaIN+RJX 5 group (p<0.05). In the histopathological examination of kidney tissues, a decrease in inflammatory cells and hydropic degeneration was detected in the groups we treated with rejuveinix. In this study, we concluded that rejuveinix has anti-inflammatory, antioxidant and renal protective effects in sepsis and sepsis-related acute kidney injury, and these effects are more pronounced at high doses.
