Assessment of whey protei̇n isolate's preventive effects on experimental model of chronic pancreatitis

Abstract (EN)

Chronic pancreatitis is an uncommon disease and it causes progressive damage in the pancreas. There are many causes of the disease as toxic, obstructive, metabolic, genetic. Oxidative stres and inflamatory mechanisms are important at pathogenesis. Exact mechanism of chronic pancreatitis is unknown and today, there is no curative treatment method. The aim of our study was assessment of protective effect of whey protein on chronic pancreatitis which has known anti-oxidative and anti-inflamatory effects. For this reason, 49 male, sprague dawley rats divided into 7 groups: 1. Group: Whey protein isolate was given orally on alternate days. Sacrified at day 14. 2. Group: Pancreatitis model formed by DBTC and sacrified at day 14. 3. Group: Pancreatitis model formed by DBTC and whey protein isolate was given orally on alternate days. Sacrified at day 14. 4. Group: Rats were accounted of control. Sacrified at day 28. 5. Group: Whey protein isolate was given orally on alternate days. Sacrified at day 28. 6. Group: Pancreatitis model formed by DBTC and sacrified at day 28. 7. Group: Pancreatitis model formed by DBTC and whey protein isolate was given orally on alternate days. Sacrified at day 28. At the end of the study, blood samples for biochemical tests and MDA and pancreas tissue samples for histopahological tests, NFκB, IL-6, TGF-β, TNF-α PCR tests, were taken. Biochemical parameters as AST, ALT, amilase, lipase; oxidative stres marker MDA; NFκB and TGF-β which are important for chronic pancreatitis pathogenesis were improved in whey protein treatment group when compared with day 28 pancreatitis group and these improvements were statistically significant (p<0.05). In conclusion, our data revealed that whey protein could probably have protective effect on chronic pancreatitis. It can be thought that whey protein shows the protective effect by anti-oxidative and anti-inflamatory features because of improvements in MDA, NFκB ve TGF-β.
