Abstract (EN)
In this study, total 56 pieces of Rhizobium spp. isolations which were previously isolated from wild leguminous plant existing in the central and the disricts of Kırşehir province (Kaman, Mucur, Akpınar, Akçakent, Çiçekdağı, Boztepe). For this purpose, morphological physiological and biochemical tests to isolate the PHB-producing capabilities Nile Blue A is designated with 43 pieces of isolates has been found to produce PHB. 5 of these 43 isolates were selected and their PHB producing yields with R. leguminosarum IFO 14778 control strain together with 0.5, 1 and 1.5 percent concentrations of molasses at the end of the 96 hour incubation period in spectrophotometric method were studied. R. leguminosarum IFO 14778 strain used cell dry weight 0.5 percent of the highest yield PHB according to molasses concentration has been identified as 24.23 percent. PHB yields of Rhizobium spp. isolates according to the cell dry weight was found between 7.30 percent and 72.58 percent in different molasses concentration. According to cell dry weights in different maximum PHB yields was identified, at HR 38-1a isolate, as 72.58 percent in 1 percent molasses concentration. In general in all isolates (exceptfor HR 41-1 isolate in 1 percent molasses concentration) a higher rate yield has been obtained than control strain. In addition, in order to identfy the siderophore producing abilities of Rhizobium spp. isolates Chrome Azurol Sulphate (CAS) Agar was used and it was determined that 50 of 56 isolates in total can produce siderophore. As a result of this study, isolates obtained from wild legume plant were found to have higher PHB yields and these can be used in bioplastic in the future, and in these context it is believed to contribute to the regulation of the ecosystem balance.
