Abstract (EN)
Olive is a tree that is specific to Mediterranean basin and one of the typical plants of the climate and has high values on fields like health, food, cosmetics and our country bears the production in significant amounts. microRNAs are 18-22 nucleotide long non-coding, single-stranded RNA sequences that regulates gene expression on post-transcriptional level and are known to be involved in various biological processes such as stress tolerance and development. In this study samples were taken on July and November from wild olive tree's pedicel and leaf tissues and their RNA extracted to identify conserved and novel miRNAs bioinformatically on the RNA sequence and small RNA deep-seq and novel and conserved miRNAs were obtained via bioinformatics. As a result 20 conserved and 23 novel miRNAs were detected. Gene ontologies of target mRNAs were done via Blast2GO and the expression levels of miRNAs and their target genes were quantified by qRT-PCR. 5 out of 7 miRNA/target gene relationship were verified.
