Application of plsenta-derived extra-embryonic mesenchymal stem cells in artifical organ manufacturing via 3-D (dimensional) cell culture

Mesenchymal stem cells are the most recent hope as the novel treatment modality that can be used in a wide clinical utility of cellular therapies and artificial organ production, due to the lack of host immunological response, easy to obtain and without any opposite ethical issues. More than the increase in artificial tissue studies, lack of prior research studies on the topic that describes the whole process from stem cells to artificial tissue production, and the lack of available and/or reliable data in Turkish literature, attract the attention. Currently, there are no completely artificial tissues that can be applicable for clinical routine. In the light of this these facts, the data collected within this thesis focus on the assessment of extra-embryonic mesenchymal stem cells isolated from placenta treated as medical waste, 3-dimensional cell culture processes in the rotatory cell culture system (RCCS), histological evaluations, and all the related optimization studies with the modifications required as a whole process for artificial tissue production. All the experiments in this thesis was carried out within the infrastructure of Cukurova University Adana Genetic Diseases Diagnosis and Treatment Application Research Center (CU AGENTEM). All the studies conducted in this thesis includes the discussion and the optimization of each step for artificial tissue production from extra-embryonic mesenchymal stem cells. Thus, the findings from this thesis study make several additional contribution including; being the first study on the field in Turkish literature and serving as a base for future studies for its clinical utility.
