Abstract (EN)
Aim: This thesis aims to synthesize a Schiff base ligand and its complexes with different transition metals, elucidate their structural properties, and evaluate their biological effects, particularly on the breast cancer cell line (MCF-7), under in vitro conditions. Material and Method: Structural characterization of the synthesized ligand and metal complexes was performed using techniques such as elemental analysis, FT-IR, NMR, UV-Vis., XRD, and SEM. Biological activity evaluations were carried out under cell culture conditions, and the effects of the compounds on cell viability were determined using the MTT method. In addition, molecular docking studies were conducted to investigate the possible binding modes and interaction properties of the CuL2 compound with acetylcholinesterase and butyrylcholinesterase enzymes. Results: Schiff base ligand was synthesized by the condensation reaction of 4-methoxy-2-hydroxybenzaldehyde, a salicylaldehyde derivative, with 2-amino-4-chlorophenol; metal complexes were prepared by reacting the resulting ligand with metal acetate salts under appropriate conditions. The structures of all synthesized compounds were elucidated using instrumental analysis methods, and results consistent with proposed molecular structures were obtained. In addition, the in vitro biological activities of the compounds were determined, and their cytotoxic effects were evaluated. Conclusion: After synthesis and characterization studies, cell culture studies in the MCF-7 cell line revealed that Schiff base metal complexes exhibited significantly stronger cytotoxic effects on cells compared to the free ligand. These findings support the evaluation of metal-containing Schiff base complexes as potential anticancer agents, and indicate that their therapeutic efficacy should be investigated in detail through further in vivo studies. 2026, 72 Pages Keywords: Aminophenol, Antiproliferative Activity, Ligand, Metal Complex, Schiff Base
