Abstract (EN)

Benzothiophenes have begun to form the subject of projects that aim to test their potential to be used as a drug, recently. In the first part of this study, antioxidant capacities of two benzothiophene derivatives, whose not being subjected in another project was detected by Reaxys®, were determined by three different methods: ABTS, DPPH and Galvanoxil. In the second step, the possible inhibition / activation effects of these derivatives on GSTM2-2 isozyme were investigated. Both of the benzothiophene derivatives showed very low antioxidant capacities compared to the standard antioxidants tested. As a result of the ABTS procedure for derivative-1, the EC50 value was calculated as 42.52 μM. However, in the experiments performed for the other two methods, the % RSA values were not high enough to calculate the EC50. The EC50 value could not be reached in any of the tests on Derivative-2. The value of Vmax for derivative-1 was reduced but the Km value was not changed. SigmaPlot software predicted the full noncompetitive inhibition model. Derivative-2 showed activation at low concentrations but inhibition over 5μM final concentration. The software stated this effect to be either full competitive or full noncompetitive inhibition; a possible mixed-type model. Keywords: ABTS, Benzothiophen, DPPH, Galvinoxyl, GSTM2, Inhibition model
