Daha sonra dolNew design psychoactive (NDP) substances have recently begun to be substituted for traditional illicit substances as drugs of abuse. Despite the increasing use of NDP, data on human metabolism are not adequately available. Since the use of these substances in the world started in 2009, their pharmacological and toxicological profiles are not yet known. Existing studies show that they have stronger effects compared to other illicit substances and that their metabolites maintain affinity for CB1 receptors in the CNS. NDP substances turn into different metabolites after entering the body. In order to detect these metabolites in biological samples, reference standards of these metabolites are needed. However, reference standard substances of the metabolites of 5F-NPB-22 are not commercially available in the world yet. In this study, samples collected after 3 hours of incubation of 5F-NPB-22 in THLE-2 human liver cell line were analyzed by high resolution mass spectrometry (LC/QTOF MS). In order to determine the metabolites in the obtained chromatogram, a total of three main metabolites were determined by scanning with the software supported database. These metabolites are ethyl ester, ester hydrolysis and methyl ester product. Since the studies on the cytotoxic effects of newly designed psychoactive substances in the human liver cell line are scarce, in this study, the anti-proliferative effect of 5F-NPB-22 on the cell was determined using the xCELLigence DP Real Time Cell Analyzer. According to the analysis result, the IC50 value for the 5F-NPB-22 substance was calculated as 66 µM. As a result of the findings obtained in this study, it has been shown that the determination of substance intake and the cytotoxic effects of 5F-NPB-22 by means of in vitro metabolism studies will provide important data to clinical and forensic laboratories. durulacaktır
