Investigation of oxidative stress in adipose tissue of brain derived neurotrophic factor heterozygous mice fed by high sucrose diet

It has known that overconsumption of sucrose content diets lead to metabolic dysfunctions such as insulin resistance, hyperglycemia and obesity. Oxidative stress increases with disruption of oxidant-antioxidant balance in adipose tissue. Adipose tissues have different adipokine profile because of its heterogeneity and these functions have change with oxidative stress. BDNF is a neurotrophin that synthesize in energy metabolism related regions such as liver, muscle, brain and adipose tissue. BDNF reduces oxidative stress in neurons by some pathways. In our study, BDNF's effects were investigated on oxidative stress in epididymal and subcutaneous adipose tissues of sucrose content diet fed transgenic mice. Four groups were constituted with standard and high sucrose contained diet fed wild type and BDNF (+/-) mice for four months. Serum 4-HNE, 3-NT, leptin, resistin, adiponectin, insulin and BDNF levels were measured by ELISA method. SOD, CAT, GPx activities and MDA levels were measured in protein fractions that obtained from subcutaneous and epididimal adipose tissues. In these tissues SOD2, GPx3 and CAT expressions were determined by using SYBR Green I dye in RT-PCR. While leptin, 4-HNE and 3-NT levels were found high, adiponectin and BDNF levels were found low in high sucrose diet fed transgenic mice. MDA levels were found low in subcutaneous adipose tissue of high sucrose diet fed transgenic mice. SOD, CAT and GPx activities were found high in subcutaneous adipose tissues of all transgenic mice. SOD2 expressions were found low in epididymal adipose tissue of both of two types of diet fed transgenic mice. CAT expressions were found low in subcutaneous adipose tissue of both of two types of diet fed transgenic mice. However, only GPx3 expressions were found low in that tissue of standard diet fed transgenic mice (p<0.05). It can be mentioned that BDNF may have a protective effect against oxidative stress in epididymal adipose tissue but our data were insufficient in subcutaneous adipose tissue for that evaluation.
