Effects of endothelial nitric oxide synthase gene t-786c polymorphism on clinical and biochemical parameters in patients with acute coronary syndrome and chronic coronary artery disease

Abstract (EN)

Nitric oxide (NO) is synthesized from L-arginine by the nitric oxide synthase (NOS) enzyme. NOS has three isforms which are nNOS (neuronal), iNOS (inducable) and eNOS (endothelial). eNOS gene T-786C polymorphism has been shown to have increased predisposition to various heart-vessel diseases such as; heart failure (2,3), coronary spasm (9, 10, 11, 12), myocardial infarction (10, 13) and atherosclerosis (14, 15, 16).The aim of our study was to determine the effects of eNOS gene T-786C polymorphism over disease generation and biochemical parameters in patients with acute coronary syndrome (ACS) and chronic coronary artery disease (CCAD). For this purpose 10 patients with ACS, 20 patients with CCAD and 31 healthy individuals proven to have normal coronaries with angiography were analyzed. eNOS T-786C genotypes were determined with polymerase chain reaction (PCR) and restriction fragment length polymorphism (RFLP) methods.The frequencies of eNOS T-786C genotypes were found to be 10% TT, 40% CT, 50% CC for the ACS group; 75% TT, 20% CT, 5% CC for the CCAD group, and 67.7% TT, 25.8% CT, 6.5% CC for the control group. The higher frequency of C/C genotype in the ACS group compared to controls suggests that CC genotype may play role in ACS generation. The lack of significant difference of the T-786C genotype frequencies between CCAD and control groups shows that T-786C polymorphism has no relationship with CCAD. Furthermore, T-786C genotypes have no effect over left ventricule hypertrophy and serum lipids.
