Analysis of foxo gen family variants in alcohol addicted,weak and weight increased individuals

Although the harmful effects of alcohol use, which has been in human life for thousands of years, are well known, a significant portion of the world's population consumes alcohol. Alcohol addiction, which occurs as a result of excessive alcohol consumption, is a chronic and recurrent disease associated with more than 200 diseases, in which loss of control over alcohol consumption and negative behaviors in case of withdrawal is observed. In addition, it is the second most common cause of morbidity and mortality in the world after high blood pressure and smoking. For this reason, it is very important to understand the etiology of alcohol addiction and to develop new diagnosis and treatment methods. Genetic factors are known to influence the etiology of alcohol addiction. In addition, by associating various genes with alcohol addiction, it has been shown that genetic predisposition can contribute to the emergence and development of alcohol addiction. FOXO1 and FOXO3A, members of the FOXO gene family, have also been associated with alcohol. In this study, DNA was isolated from peripheral blood obtained from alcohol-dependent individuals with different Body Mass indices and controls in order to determine the relationship between FOXO1 and FOXO3A gene variants with alcohol addiction. Then, by determining the genotype frequencies and allele frequencies with the PCR-RFLP method, statistical analysis were performed. As a result of the experiments, it was determined that the GG genotype increased the susceptibility to alcohol addiction for the rs3900833 polymorphism in the FOXO1 gene (p = 0.03), and the CC genotype for the rs4581585 polymorphism increased the susceptibility (p = 0.03). But, it has not been any significant relationship between rs4966936 and rs2253310 polymorphisms in FOXO3A gene and alcohol addiction (p>0,05).
