Abstract (EN)

In the present study, Cu(II) ions were enriched by solid phase extraction method and determined by Flame Atomic Absorption Spectroscopy. For this aim, Anoxybacillus sp. SO-B1 bacterium, immobilized in Amberlite XAD-16 resin, was filled in a mini column and this was used as the solid phase. In order to optimize the method, effect of some analytical parameters, such as pH of the solution, flow rate of the solution and the eluent, type, concentration and volume of the eluent, volume of the sample solution on recovery yield of Cu(II) ions were investigated. Furhermore, adsorption capacity of the biosorbent was studied for copper. Experimental studies was caried out at pH 7.0 and eluation was performed with 5 mL of 1 M HCl. Quantitative values were obtained in adsorption of Cu(II) ions and also in its eluation.In the optimum conditions, recovery yields of Cu(II) ions were found to be 100.88 ± 1.57 (N=5). In determination by AAS, limit of detection for Cu(II) was found to be 0.16 µg/L (N=10). In order to investigate applicability of the method to real samples, influence of several matrix ions Na+, K+, Mg2+, Cl- and NO3- etc. on recovery yield was studied.The developed method was applied to real samples such as Tigris River water, Elazığ Maden Stream water, Hazar Lake water and tap water for determination of Cu(II) ions. Accuracy of the method was checked by analysing standard reference material.
