Investigation of antioxidant and antimicrobial properties of extract of Astragalus diphtherites Fenzl var. diphtherites ve Astragalus gymnalocepias Rech (fabaceae) taxa preparead by solvent having different polarity

Present work reports in vitro antimicrobial activity and antioxidant characteristics, hydroxyl radical scavenging activity, reducing power, metal chelating activities, 2,2-Diphenyl?1-picryl-hydrazil (DPPH) radical scavenging activity, total phenolic and total flavonoid content of hexane, ethyl acetat, acetone and methanol extracts obtained in the supernatant of stem and roots of Astragalus diphtheries Fenzl var. diphtherites and Astragalus gymnalocepias Rech (Fabaceae) taxa.The highest total phenolic content was obtained from the stem (76.1±0.9 µg GAE/mg) in the methanol and the root (48.02±1.5 µg GAE/mg) in the aceton extracts of A. diphtheries var. diphtherites. For A. gymnalocepias, the highest phenolic content was obtained from methanol extract of stem (54.66±02.25 µg GAE/mg) and ethyl acetat extract of root (35.83±1.75 µg GAE/mg).The flavonoid content of extracts calculated as quercetin equivalent. The highest total flavonoid content of A. diphtheries var. diphtherites was measured in the stem (42.20±0.46 µg QUE/mg) with acetone, and in root (4.23±0.26 µg QUE/mg) with ethyl acetate extract. For A. gymnalocepias, the total flavonoid content was obtained from stem (80.15±0.33 µg QUE/mg) in the acetone, and the root (14.01±0.10 µg QUE/mg) in the ethyl acetate extract.For A. diphtheries var. diphtherites, the highest DPPH radical scavenging activity was measured in the stem (79.01±0.74%) extract with methanol, and in the root extract (91.55±0.24%) with aceton extract, respectively. In the case of A. gymnalocepias, this activity was 86.83±2.24% with methanol and 84.10±1.99% with ethyl acetate fraction.The highest reducing power activity was shown with the stem and the root extracts of A. diphtheries var. diphtherites and A. gymnalocepias plants with acetone and ethyl acetate extract respectively.Ethylene Diamine Tetra Acetate (EDTA) was used as standarts for the metal chelating activity. The highest chelating activity was determined in the stem (74.35±1.51%) with methanol and the root (62.42±0.73%) with hexane extracts of A. diphtheries var. diphtherites. In the case of A. gymnalocepias, this activity was 89.63±0.58% and 76.41±0.90% with methanol extracts.Dimethyl sulfoxide (DMSO) was used as standard for DPPH radical scavenging activity. The highest DPPH radical scavenging activity was determined in the stem and the root with ethyl acetate extracts of A. diphtheries var. diphtherites and A. gymnalocepias.The antimicrobial activities for hexane, ethyl acetate, acetone and methanol extracts of the stem and root of A. diphtheries var. diphtherites and A. gymnalocepias were tested by disc diffusion method. Escherichia coli ATCC 25922, Staphylococcus aureus ATCC 25923, Streptococcus pyogenes ATCC 19615 Pseudomonas aeruginosa ATCC 27853, Bacillus subtilis ATCC 11774 together with the Candida albicans yeast ATCC 10231 and the standart antibiotics such as Eritromisin, Amoxicillin, Ofloksasin, Netilmisin and Amfoterisin B and 80 and 160 ? g/paper disc plant extracts were evaluated using. As a result of the study, it was observed that there is inhibition only the stem extracts of A. gymnalocepias with acetone and methanol on the growth of Streptococcus pyogenes.
