Abstract (EN)

There is no rational treatment of epilepsy, which affects more than 50 million people in the world and is defined as a neurological disorder characterized by recurrent seizures and loss of consciousness as a result of excessive activation of neurons. The main strategy in the treatment of epilepsy is the prevention of abnormal electrical discharges. There are many commercialized drugs for this purpose. Compounds including pyrazole and isoquinoline ring having a wide variety of biological and pharmacological effects are known to exhibit anticonvulsant activity by inhibiting these discharges separately. In this study, it is aimed to synthesize new heterocyclic compounds which will contain one or both of these two active rings and to investigate the anticonvulsant activities. For this purpose, the reaction of the 3,5-diphenyl-pyrazole compound synthesized as starting material with different 2-bromo-aryl-ethanone derivatives resulted in the synthesis of 3 new 2-pyrazolo-aryl-ethanone derivatives. 3 new pyrazolo-isoquinoline derivatives were synthesized by ring closure due to dehydration of these synthesized compounds in a strongly acidic medium. Studies of the in vivo anticonvulsant activities of these compounds, whose structure was illuminated by spectral data, were investigated by the behavioral effects of akut epilepsy model. It is seen that the synthesized compounds have no behavioral anticonvulsant effects in the acute PTZ model. However, if necessary, these substances may be tested in the chronic epilepsy model or in other experimental epilepsy models. Keywords: anticonvulsant activity, epilepsy, isoquinoline, pyrazole, pyrazolo-isoquinoline.
