Abstract (EN)

Cisplatin is an essential element of chemotheraphy protocols which plays a role in the treatment of a few tumor seen in adulthood and many childhood tumors. Cisplatin is indicated as partial gonadotoxicity which has the adverse affects in many systems as shown in studies. Nephorotoxicity, neuropathy, ototoxicity and gonadotoxicity are clinically important dose-limiting side effects that limit the use of Cisplatin. After Cisplatin entering inside the cell, it shows the cytotoxic effect in cell leading to apoptosis, necrosis, oxidative stress, fibrogenesis, inflammation, hypoxia and mitochondrial damage by activating a multi-signal transduction pathway.The purpose of this study; known the effects of gonadotoxicity which is caused by cisplatin, we want to determine the effercts in rats tuba fallopians and present the protective effect of Acetyl L-Carnitine which is determined against these effects.An examination of these effects, hematoxylin-eosin(H&E) staining was applied for morfometric and histologic analysis and determine the apoptotic effects by by using Caspase 3, 8, 9 and TUNEL methods were applied to identify DNA fragmentation.When the mean weight was evaluated before and after the experiment, weight loss was obsereved in Cisplatin and pre-treatment group. However in Cisplatin Groups weight loss was greater than pre-treatment group.In Cisplatin group, tuba uterina wall thickness and the thickness of the epithelium had slimmed. But the avarage thickness of the wall and epithelium in Acetyl L-Carnitine+Cisplatin pre-treated group thicker than Cisplatin group.During histological examinations, the presence of intraepithelial lymphocytic infiltration was observed in Acelty L-Carnitine+Cisplatin groups. Subepithelial hemoragy, which was observed in Cisplatin group, was not detected in Acetyl L-Carnitine+Cisplatin group.In fallopian tube epithelial, a large number of cells DNA fragmentation was observed. It was observed that Acetyl L-Carnitine+Cisplatin pre-treatment practice prevents the fragmentation. The result of Caspase 8, 9 and 3 immunohistochemistry which is caused by Cisplatin in order to demonstrate apototik pathway, showed that apoptosis was triggered with two pathways as instrinsic and extrinsic.Acetyl L-Carnitine+Cisplatin pre-treatment application failed to prevent apoptosis triggered by instrinsic pathways but prevented the apoptosis triggered by extrinsic pathways over Caspase 8.As a result of this study that occurs toxic effects of Cisplatin application on tuba uterina and Acetyl L-Carnitine prevent to histomorphologic and apoptotic effects which is applied prophylactically.
