Investigation of the effect of Mangiferin in an experimental ovarian ischemia-reperfusion injury model

Abstract (EN)

Objective: This study aimed to evaluate the potential antioxidant, anti-inflammatory, and protective effects of Mangiferin in an ovarian ischemia-reperfusion (IR) injury model. Materials and Methods: A total of 32 female Sprague-Dawley rats were divided into four groups: Control, Mangiferin, IR, and IR + Mangiferin. The IR model was established by clamping the bilateral ovarian pedicles for 3 hours followed by 3 hours of reperfusion. Mangiferin was administered intraperitoneally at a dose of 20 mg/kg for 15 consecutive days after reperfusion. Tissue MDA levels, serum AMH concentrations, follicle counts, histopathological scores, and TNF-α and NF-κB expressions were assessed. Results: In the IR group, MDA level was 60 nmol/g, TNF-α expression was 64.9%, NF-κB expression was 69.5%, and AMH level decreased to 1.2 ng/mL. A reduction in primordial and primary follicle counts was observed, accompanied by histological findings such as edema, hemorrhage, and inflammatory cell infiltration. In the IR + Mangiferin group, MDA level decreased to 24 nmol/g, TNF-α to 44.8%, and NF-κB to 41.4%, while AMH level increased to 4.6 ng/mL. Follicle counts improved and histological damage findings were significantly reduced. Conclusion: Mangiferin effectively attenuated oxidative stress and inflammatory response in the ovarian IR model and contributed to the preservation of follicular structures and ovarian reserve.
