Abstract (EN)
The infections which are caused by dermatophites in humans and animals are called dermatophytosis and tinea. The gold standart in laboratory diagnosis of dermatophytosis are direct microscobic analysis and culturing fungi. The sensitivity of direct microscobic investigation is higher than culturing however the pseudo negative result and the impossibility of species assignment are the disadvantages reported. The genus and type of dermatophytes are determined by classical and moleculary identification method. In the study presented SGA; the fundamental medium for laboratuary diagnosis of dermatophytes and MBLA; which induces sporulation and pigment formation and helps detecting dermatophytes in species level of diagnosis were used syncronously by plantation and comparison of nutrition performance and the results that were obtained were aimed to discussion. In this study 331 samples have been researched which were collected from hairy skin, bald skin and nails of 257 patients who were hospitalised in different policlinic services with dermatophytosis suspicion whom applied to Çukurova University Dermatology Department between september 2014- january 2015. 114 samples of 81 people were included in this study which had positively resulted from either one of direct microscopy or culturing. In this study it was understood that MBLA 55(%21.4) has shown better performance than SGA (%19.1) and it should be used in routine laboratuary practice. Almost all kinds of dermatophytes were spored better in MBLA and were better identified by the the pigment which was produced. Being time consuming to be prepared and being blurry in medium have been discovered to be the disadvantages of MBLA. It was found to get more positive results comparing to the other 2 mediums by being easier to apply and not requiring experience, being cheaper, and being able to be analysed with direct microscopy. In this study for the first time, in addition to the colonial and morphological features, the dermatophyte flora of a city in our country were determined with classical methods together with ITS sequence analysis on limited amount of samples. The species that were detected from the 63 isolates identified in this study were: Trichophyton spp. (n=45), T. rubrum (n=10), T. interdigitale (n=4), E. floccosum (n=2), T. violaceum (n=1) ve M. canis (n=1) Tinea Unguim has higher prevalance in men (p=0.006), and the prevalance of Tinea unguium is increased by age and this increase makes a difference. For more realistic results in the labratuary diagnosis of dermatophytosis at least two mediums used together are considered to be useful.
